British Columbia man cured of rare disease in world-first for new gene-editing technology

Patient Background

Ty Sperle, a 19-year-old from Kelowna, B.C., was diagnosed with CGD around age five. This immune system disorder makes patients highly susceptible to severe, potentially fatal infections. Treated for over a decade by Dr. Stuart Turvey at B.C. Children’s Hospital, Sperle had no suitable stem cell donor for traditional therapy.

The Treatment

Sperle received “prime editing,” a breakthrough gene-editing technology from U.S.-based Prime Medicine. Doctors extracted his stem cells, corrected the specific genetic “spelling mistake” causing CGD, and reinserted them; the fixed cells then multiplied and overrode faulty ones without rejection risk. The procedure occurred at Montreal’s Sainte-Justine university hospital, marking the first known cure via this method, as detailed in the New England Journal of Medicine last December.

Sperle’s Reaction

Sperle described an “insane shock” and overwhelming happiness upon learning he was cured, ending his daily medication regimen and constant infection fears. Now a second-year science student at UBC Okanagan, he endured extreme stress during the transplant but trusted his doctors; his mother cried tears of joy when he shared the news. He hopes this inspires others with genetic diseases.

Significance

Dr. Turvey called it a “dream come true,” highlighting how gene editing translates genomic discoveries into patient care; rare diseases like CGD affect about one in three B.C. Children’s Hospital patients collectively. This autologous approach avoids donor issues, paving the way for broader applications as the technology advances.
